The primary objectives involve navigating through seven distinct locations, including Egypt, the Amazon, Italy, Camelot, the USA, Transylvania, and the Moon. In each area, players must jump on and destroy alien eggs and hatched aliens while also finding and releasing imprisoned heroes, such as Indiana Jones and Luke Skywalker. The gameplay focuses on platforming movement and avoiding environmental hazards, with each location featuring unique enemies and backdrops, like mummies in Egypt or classic horror characters in Transylvania.

Progression is linear through the series of themed stages, with players needing to clear enemies and complete rescue objectives to advance. A key gameplay system involves collecting power-ups, specifically an "S" icon that temporarily transforms Luigi into Superman, granting him invulnerability to enemy contact. Without this power-up, any touch from an enemy causes damage.
